Etymology dictionaryによれば、1620s, "represent in bodily form," from assimilated form of Latin in- "into, in" + persona "person." Sense of "assume the person or character of" is first recorded 1715 とあります。
詳しい説明はありませんが、ラテン語由来でしょうか？1715年に「人のふりをする」と言う意味が登場したらしいです。
意味
Oxford learner's dictionariesによれば、impersonate somebodyの形で、to pretend to be somebody in order to trick people or to entertain themと説明があります。つまり、「～になりすます」、「～のふりをする」です。

警視庁も、公務員になりすまして、早期の予防接種であるとして代金を支払うよう高齢者を騙そうとする不審な発信者に対して警告するチラシを配っている。
conは、 con somebody (into doing something)の形で使われます。「（人）をだまして～させる」の意味です。前置詞intoの後には、その結末が来ます。
